RESTRICTION ENDONUCLEASE ANALYSIS OF HYPHANTRIA CUNEA NPV

Abstract: Viral DNA was isolated directly from the nuclear polyhedrosis virus of Hyphantria cunea. Viral DNA showed a single band on the agarose gel. Digestion of HcNPV-DNA with EcoR Ⅰ, Pst Ⅰ, Bgl Ⅱ, and PVU Ⅱ resulted in 19, 22. 18 and 22 fragments on agarose gel electrophoresis respectively. The molecular weights of the fragments are determined with DNA-EcoRI and DNA-Hind Ⅲ fragments as reference, and using this method the mean molecular weight of HcNPV-DNA is calculated to be about 94.044×106 daltons.
